The included cell line overexpresses PTAFR alongside endogenous high levels of a promiscuous G-protein, coupling the receptor to the calcium signaling pathway. Upon activation of PTAFR, the resulting calcium activity can be fluorescently assayed. The cell line overexpresses ProLink-tagged fusion of the KDR kinase domain with transmembrane and extracellular ligand binding domain of EGFR; and also an EA-tagged SH2 domain protein. Upon ligand induced receptor activation, cross-phosphorylation of the receptor leads to SH2-EA recruitment and production of EFC signal.
